If we don’t get kits in time, then just do the following:1. Read the appropriate content in the textbook regarding pH and its measurement. 2. Extract pH indicator from red cabbage. You will need to buy red cabbage.3. Buy a paper coffee-maker filter if you don’t get your kit in time.4. Make pH paper by soaking filter paper with red cabbage liquid and letting it dry.5. Dip your filter paper in products around the house and noting any color changes.6. Repeating this with commercial pH paper, if you have any. Exercise 2 - Preparation of pH Indicators from Anthocyanins In this exercise, you will extract the anthocyanin pigment from red cabbage to create pH strips. Note: It is recommended that you wear an apron since the dye from the cabbage will stain so be careful to not damage your clothing or carpet! Procedure1. Remove the outer leaves of the red cabbage.2. Place the red cabbage on the cutting board. Using the knife, chop the cabbage into pieces, as shown in Figure 8. Chopping the cabbage.1. Using a measuring cup, measure 1 cup of tap water.2. Bring the cup of tap water to a boil. You can microwave it in a heat-safe bowl or boil it in a tea kettle.3. When the water reaches a boil, use an oven mitt to hold the container, and (if it is not already in a heat-safe bowl) carefully transfer the boiling water into a heat-safe bowl. 44. Add 1 cup of chopped red cabbage into the heat-safe bowl with the boiling water.5. Use a spoon to ensure that the red cabbage is fully immersed in the water, as shown in Figure 9. Figure 9. Steeping the cabbage.1. Allow the red cabbage to steep in the water for 20 minutes. Note: This is not the "drying time" for this exercise.1. After the red cabbage has steeped in the water for 20 minutes, use the spoon to mix the cabbage and water. This will allow the deep purple color to be evenly distributed throughout the bowl. CAUTION! This dye will stain so be careful to not damage your clothing or carpet.1. Cut the filter paper into 4 equal-sized pieces. If you did not receive filter paper, buy and cut up a paper coffee-maker filter.2. Submerge 1 cut piece of filter paper into the bowl of red cabbage and water. The filter paper must be fully submerged in the bowl, as shown in Figure 10. Allow the filter paper to absorb the colored water for 5 minutes. 4. Add 1 cup of chopped red cabbage into the heat-safe bowl with the boiling water.5. Use a spoon to ensure that the red cabbage is fully immersed in the water, as shown in Figure 9. Figure 9. Wash and dry the bowl.4. Allow the filter paper to dry for approximately 4 hours before proceeding on to the next step. Note: This is the "drying time" for this exercise.1. When the filter paper is dry, cut the paper into strips, approximately 0.5 cm × 5.0 cm. Be sure that the width of the strip does not exceed 0.5 cm; otherwise the strip may not fit into the well plate. See Figure 11. Figure 11. Strips of homemade pH paper. 1. Place the cut filter paper aside for use in Exercise 3. These cut filter papers will be referred to as "Homemade pH paper" for the remainder of this lab.Exercise 3 - Testing Common Household Material sin this exercise, you will test the acidity/basicity of common household materials, and compare commercially prepared pH strips to the homemade pH strips you prepared in Exercise 2.Note: It is important that you conduct this exercise properly:•Review the safety materials and wear gloves and goggles when working with chemicals.•Read the entire exercise before you begin.•Take time to organize the materials you will need and set aside a safe work space in which to complete the exercise.Procedure1. Gather 9 household liquid items to test, such as shampoo, soap, lemon juice, soda, milk, and cleaning supplies.2. Record these items in Data Table 4.3. Put on your safety goggles and gloves.4. Place the following items into the first 3 wells in the 24 well plate. If you don’t have the well plate, use glass or plastic cups. Rinse the cups out when after each use. Well #1: Fill halfway with 1M HCl (hydrochloric acid).well #2: Fill halfway with 0.1M NaOH (sodium hydroxide).well #3: Using a pipe, fill the well with distilled water. Using and Reading Wide-Range pH Paper1. Using a separate pipet for each item, place each household liquid item that you have selected into its corresponding well, filling each well halfway. (Just pour a little household product into a cup if you did not receive your kit or pipet.)2. Remove the pH test strips and pH color range guide from the kit. We will refer to the pH strips provided in the kit as the "commercial pH strips" for the rest of this exercise. (Ignore testing with commercial test strips if you did not get your kit.)3. Remove 6 pH test strips from the book and cut them in half, lengthwise, creating 12 half-sized strips. The half-sized strips will be referred to as "strips" for the remainder of the experiment. See Figure 12. Figure 12. Cutting the pH strips in half, lengthwise. 1. Place the pH color range guide next to the well plate, so that it is easy to see while conducting the experiment.2. Wearing gloves, pick up 1 commercial pH strip. Holding the strip at 1 end, dip the bottom of the strip into Well #1 to saturate the strip with the liquid in the well. See Figure 13. Figure 13. Record the color of the strip and its corresponding pH in Data Table 4 in the column labeled "Commercial pH Strip Color and Value."3. Repeat steps 9 through 11 for the remaining 11 samples in the plate. Use a fresh strip of pH paper for each well.4. Repeat steps 9 through 12 for the homemade pH strips you prepared in Exercise 2. A pH color guide for the homemade pH strips is shown in Figure 14. Figure 14. Homemade pH strip color guide. 1. Review the results in Data Table 4. Using both the commercial pH color range and homemade pH strip color guide, classify each substance as acidic, neutral, or basic and record these results. Data Table 4 in the column labeled "Acid/Base/Neutral?" Additionally, write a short explanation of how you decided on each classification. Cleanup:•Using the test tube cleaning brush, thoroughly wash the 24 well plate with soap and water.•Retain the unused pH papers and chemicals for use in Exercise 4.•Dispose of the used pH papers in the trash.•Return the cleaned and dried well plate to the kit for future use.
