I need my portion of an approach section in a grant proposal done by tomorrow no later than 7pm! Each aim must include the rationale, the experiment for each, the pitfalls, and expected results. AIM 1- Investigate whether Cx43 mediates Ca2+ transients. 1.1 Identify Cx43 expression in RGC’s. 1.2 Loss of function Cx43 and Ca2+ transients. 1.3 Gain of function Cx43 and Ca2+transients. Figures and citations also need to be included! The citations can just be left at the bottom of the page I will put them in a works cited myself! I have attached the grant proposal guidelines, as well as my professor's grant that he submitted as an example of what my portion needs to look like! The main experiments for each aim are as follows, immunostaining and Nestin protein in Aim 1.1, Inuteroelectroporation, ex vivo and Calcium 2+ ion imaging for Aim 1.2 and Aim 1.3. ALL EXPERIMENTS ARE DONE ON MICE

Separate the ventricular zone, subventricular zone and cortical plate by dissection. Run western blots and immunostaining for each section. Combine immunostaining with RGC fiber markers (NESTN). Identify if Cx43 is present on fibers. Cx43 should be located in all three zones as calcium transients travel throughout RGC fibers. Calcium imaging to knock down and over express Cx43. By targeting radial glia cells in developing mice embryos, immunostaining for Connexin43 will be marked with a green fluorescent protein. Micro-dissections between the pial membrane and ventricular membrane will include separation of the ventricular zone, subventricular zone and cortical plate where Connexin43 stains should be consistent along the RGC cell fiber. Consistent staining along the RGC cell fiber will potentially indicate a communication pathway using the Connexin43 protein.
